Johnny Gregorius is the grandson of Juan Gregorius, son of Didi Gregorius and brother of Mariekson Gregorius. A former member of the Dutch Antilles youth national team, he had been pitching for the Sta. Maria Pirates in the top Curacao-based league when he was signed by the ADO club in 2007 thanks to the influence of Jermain Esprit. In his debut in May, he allowed 11 runs on 6 hits and 3 walks in 3 innings against DOOR Neptunus in a 15-5 defeat. He was 0-2 with a 9.90 ERA, allowing 17 hits and 12 walks in 10 innings in the 2007 Hoofdklasse. Not invited back to Neptunus in 2007, Gregorius joined his father on the Netherlands Antilles national baseball team for the 2008 Americas Baseball Cup and tossed a 2-hit shutout in a win over the Panamanian national team. In his other start, he allowed 3 hits and no runs in 6 innings. His 2-0, 0.00 record earned him a spot on the tourney's All-Star team as he helped the Antilles clinch a spot in the 2009 Baseball World Cup. To round out his dominant work, he even went 4 for 7 with 4 runs at the plate.

Gregorius hit .338/.378/.481 with 23 runs in 18 games for the Santa Maria Pirates in 2008, helping them win the Netherlands Antilles pennant; on the mound, he was 1-1 with a save and a 4.05 ERA.

Gregorius signed with T & A San Marino for 2009; after no Dutch citizens had played in Italy for about two decades, Gregorius joined Johnny Balentina, Ivanon Coffie and Ardley Jansen in making the transition for 2009. He had a rough debut in the Italian Baseball League, allowing 3 runs in 2 relief innings against Reggio Emilia on April 11 in a 5-4 win for his club.

Gregorius played for the Antilles in the 2010 South American Games.
